Methods of identification of filamentous bacteria as a disturbing factor of wastewater treatment plants work.


The sewage treatment process using activated sludge is currently the most widely used method of biological wastewater treatment. This process is divided into two stages. One, called biochemical takes place in aeration tanks. Second, the physical takes place in a secondary clarifier. In the first stage the decisive role played by activated sludge, which is a consortium of many microorganisms, performing a variety of roles. Under appropriate conditions, they are able to efficiently remove organic compounds from the treated waste.
One group of microorganisms present in the sludge are filamentous organisms. Their presence in the activated sludge can cause such disruption to plant operation, as filamentous bulking or its foaming. Over the years, various methods have been proposed to ensure the prevention and control of such phenomena. Unfortunately, none of the currently used approaches do not give a complete guarantee eliminate them.
To identify filamentous organisms are used in various types of identification keys.
More recent solution is the molecular methods based on analysis of DNA and RNA of the bacteria. They are considered more reliable and less subject to error. For sludge application now found a method that uses unique for each type of microbial 16S rRNA. Based on 16S rRNA sequence of the known, it is possible to design probes that react with the specific sequence. They can be used for dyeing of certain bacteria. In this way it is possible to identify the bacteria by FISH. Furthermore, many types of bacteria still not been identified taxonomically. In recent times we discovered and described approx. 40 new morphotypes of filamentous bacteria. This paper provides an overview of the main groups of filamentous bacteria and the methods of identification in the activated sludge.

Furthermore, many types of bacteria HAVE still not been identified taxonomically.
